Approved systemic AAV9 comparator: onasemnogene abeparvovec (Zolgensma) label attributes
AttributeQuoted label text
Indication“ZOLGENSMA is an adeno-associated virus (AAV) vector-based gene therapy indicated for the treatment of pediatric patients less than 2 years of age with spinal muscular atrophy (SMA) with bi-allelic mutations in the survival motor neuron 1 (SMN1) gene.”
Route and schedule“For single-dose intravenous infusion only”
Dose“The recommended dosage of ZOLGENSMA is 1.1 × 10¹⁴ vector genomes (vg) per kg of body weight.”
Immunosuppression“Starting one day prior to ZOLGENSMA infusion, administer systemic corticosteroids equivalent to oral prednisolone at 1 mg/kg of body weight per day for a total of 30 days.”
Boxed warning“WARNING: SERIOUS LIVER INJURY and ACUTE LIVER FAILURE”“Cases of acute liver failure with fatal outcomes have been reported.”
Antibody testing“Perform baseline anti-AAV9 antibody testing prior to ZOLGENSMA infusion.”“The safety and efficacy of ZOLGENSMA in patients with anti-AAV9 antibody titers above 1:50 have not been evaluated.”
Liver monitoring“Continue to monitor liver function for at least 3 months after infusion, and at other times as clinically indicated.”
Platelet monitoring“Monitor platelet counts before ZOLGENSMA infusion, and at least weekly for the first month and then every other week for the second and third month or until platelet counts return to baseline.”
Thrombotic microangiopathy“Prompt attention to signs and symptoms of TMA is advised, as TMA can result in life-threatening or fatal outcomes.”
Cardiac monitoring“Consider cardiac evaluation after ZOLGENSMA infusion and consult a cardiologist as needed.”
Approved systemic AAVrh74 comparator: delandistrogene moxeparvovec (Elevidys) label attributes
AttributeQuoted label text
Indication“ELEVIDYS is an adeno-associated virus vector-based gene therapy indicated for the treatment of patients 4 years of age and older with Duchenne muscular dystrophy (DMD) who are ambulatory and have a confirmed mutation in the DMD gene.”
Route and schedule“ELEVIDYS is for single-dose intravenous infusion only.”
Dose“Recommended dosage: 10 to 70 kg: 1.33 × 10¹⁴ vector genomes (vg) per kg of body weight; 70 kg or greater: 9.31 × 10¹⁵ vg.”
Immunosuppression“One day prior to infusion, initiate a corticosteroid regimen for a minimum of 60 days.”
Boxed warning“WARNING: ACUTE SERIOUS LIVER INJURY AND ACUTE LIVER FAILURE”“Acute serious liver injury, including life-threatening and fatal acute liver failure, has occurred with ELEVIDYS.”
Antibody testing“Select patients for treatment with ELEVIDYS with anti-AAVrh74 total binding antibody titers <1:400 as measured by an FDA authorized test.”
Liver monitoring“Continue to monitor liver function weekly for the first 3 months after infusion and continue until results are unremarkable.”
Proximity to care“Instruct patients to maintain proximity to an appropriate healthcare facility, as determined by the healthcare provider, for at least 2 months following ELEVIDYS infusion.”
Cardiac monitoring“Acute, serious, life-threatening myocarditis and troponin-I elevations have been observed.”“Monitor troponin-I before ELEVIDYS infusion, and weekly for the first month after ELEVIDYS infusion.”
Infection risk“Serious infections with fatal outcomes may occur due to concomitant administration of corticosteroids, additional immunosuppressants, and ELEVIDYS.”
