Summary
AIC316-01-II-01 enrolled otherwise healthy, immunocompetent adults with genital HSV-2 infection, a population outside the labeled indication of immunocompromised adults with refractory mucocutaneous HSV infection. The regimens tested (5, 25, or 75 mg daily, or 400 mg weekly) differ from the approved regimen of 400 mg on day 1 followed by 100 mg daily. Treatment lasted 28 days, and 155 participants were analyzed in five arms of 29 to 33 participants. The primary endpoint was a virologic measure, and the investigators state that the trial was not powered to evaluate recurrences. The publication reports one serious adverse event (pancreatitis 2 weeks after treatment), and the registry results list none.
