Summary

AIC316-01-II-02 enrolled healthy, immunocompetent adults with 4 to 9 genital HSV-2 recurrences per year, a population outside the labeled indication of immunocompromised adults with refractory mucocutaneous HSV infection. The pritelivir regimen (400 mg on day 1, then 100 mg daily) matched the approved dose and was given for 28 days to suppress shedding. The sponsor terminated the trial after the FDA clinical hold of May 2013; 91 participants were randomized against 98 planned, and 56 completed both treatment periods. For the primary endpoint, the publication reports a relative risk of 0.42 (95% CI, 0.21 to 0.82) and the registry reports a risk ratio of 0.43 (95% CI, 0.30 to 0.63). The publication was corrected in January 2017.
